4-fumarylacetoacetic acid, also known as fumarylacetoacetone or (E)-4,6-dioxo-2-octenedioate, belongs to medium-chain keto acids and derivatives class of compounds. Those are keto acids with a 6 to 12 carbon atoms long side chain. Thus, 4-fumarylacetoacetic acid is considered to be a fatty acid lipid molecule. 4-fumarylacetoacetic acid is slightly soluble (in water) and a weakly acidic compound (based on its pKa). 4-fumarylacetoacetic acid can be found in a number of food items such as fenugreek, lime, longan, and custard apple, which makes 4-fumarylacetoacetic acid a potential biomarker for the consumption of these food products. In humans, 4-fumarylacetoacetic acid is involved in few metabolic pathways, which include disulfiram action pathway, phenylalanine and tyrosine metabolism, and tyrosine metabolism. 4-fumarylacetoacetic acid is also involved in several metabolic disorders, some of which include alkaptonuria, tyrosinemia type 3 (TYRO3), phenylketonuria, and tyrosinemia type I. Fumarylacetoacetic acid (fumarylacetoacetate) is an intermediate in the metabolism of tyrosine. It is formed through the conversion of maleylacetoacetate into fumarylacetoacetate by the enzyme maleylacetoacetate isomerase .
